Background and Aim: The hormone receptor status in breast cancer has been pivotal in determining the likelihood of response to hormonal manipulation. Tumors which are both estrogen and progesterone receptor positive are much more likely to respond to anti-hormone therapy than negative tumors. There is well-established similarity between breast tissue and salivary glands. The aim of this study was to evaluate the progesterone receptor expression in pleomorphic adenoma and adenoid cystic carcinoma of salivary glands. Materials and Methods: In this descriptive study, immunohistochemical staining with progesterone antibody was performed on 14 pleomorphic adenoma (PA) and 15 adenoid cystic carcinoma (ACC) paraffin blocks. The percentage of positive cells was determined using an eye piece graticule. Immunoreactivity was categorized as either positive (reactivity more than 5%) or negative (reactivity less than 5%). In addition the existence of progesterone receptor in tumor cells, stromal cells (fibroblasts), inflammatory cells and salivary glands around tumors was evaluated. Data were analyzed with T and Mann Whitney U tests with p<0.05 as the limit of significance. Results: Immunohistochemical staining for progesterone receptor was negative in 15 ACC and 13 PA. Only one case of PA showed immunoreactivity for progesterone receptor. Also, 12 normal salivary glands around tumor were positive. Inflammatory cells, endothelial cells and fibroblasts did not show immunoreactivity in most cases. Conclusion: The results indicate the lack of progesterone receptor expression in ACC and PA of salivary glands.

Statement of Problem: Differentiation of dentigerous cyst from unicystic ameloblastoma, discovering any initial ameloblastic changes in lining epithelium of dentigerous cyst at early stage, and differentiation between hyperplastic odontogenic epithelium in fibrous capsule of dentigerous cyst from ameloblastic proliferation, need to an accurate and reliable technique.Purpose: The aim of this study was to determine and compare Ki-67 immunoreactivity in various locations of the epithelium of Dentigerous cyst and Unicystic Ameloblastoma.Materials and Methods: In this historical Cohort study, 15 cases of dentigerous cyst and 9 cases of unicystic ameloblastoma were selected. Immunohistochemistry staining was performed by MIB-1 (murine monoclonal antibody against Ki-67). The stained nucleous were counted in basal and suprabasal layer of lining epithelium of both lesions in 1000 epithelial cells. Finally, the percentage of positive cells (presented as labeling index) was calculated. t- Student test was used to analyze the related data. Results: Ki-67 (LI) in basal layer of Dentigerous cyst (2.59 1.66) and Unicystic Ameloblastoma (3.76 79) had no significant differences, but Ki-67 (LI) in suprabasal layer of unicystic ameloblastoma (2.15 0.69) was significantly higher than dentigerous cyst (0.77 0.55) P=0.003).The difference between the average numbers of positive cells for Ki-67 (LI) in these two lesions was statistically significant (P<0.05) and it was higher in Unicystic Ameloblastoma than Dentigerous cyst. Conclusion: Based on the findings of this study, it is suggested that Ki-67 (LI) in suprabasal layer or throughout the epithelium can be considered as a useful marker for differential diagnosis between dentigerous cyst and unicystic ameloblastoma.

Abstract:

Background: The p53 gene mutation is closely related to carcinogenesis in most malignant diseases. The main function of wild p53 protein is to maintain the integrity of genes by detecting mutations and preventing the division of cells with damaged DNA. The mutated form of p53 protein is overexpressed due to an extended half-life and can be easily detected by immunohistochemistry. Objective: To estimate the frequency of p53 protein overexpression in colorectal carcinoma and its correlation with some clinicopathologic variables. Methods: One hundred paraffin-preserved colorectal carcinoma samples were collected randomly from patients undergoing tumor resection from April 1995 through April 2001 in Omid Hospital, affiliated to Mashhad University of Medical Sciences, Mashhad, Iran. The overexpression of p53 protein was studied using a monoclonal antibody (clone DO-7; Dako). The number of cells stained were classified semiquantitatively as (–): <5% positive cells, (+): 5 – 25% positive cells, (++): 25 – 75% positive cells, and (+++): >75% positive cells. Clinicopathologic data including gender, age, tumor location, histologic type, and stage (Astler-Coller) were collected from the files maintained at the Department of Pathology. The correlation between p53 protein overexpression and each variable was evaluated using Chi-square analysis. Results: p53 staining was positive in 59 of 100 specimens. Out of these 100 specimens, 16 were weekly (+), 16 moderately (++), and 27 intensely (+++) positive for p53 protein overexpression. There was no significant correlation between p53 staining and gender (P=0.34), age (< 40 vs. ≥ 40 yr; P=0.74), site of tumor (right vs. left colon and rectum; P=0.26), pathologic type (mucinous vs. nonmucinous; P=0.63), and stage of the disease (P=0.12).Conclusion: Considering the p53 protein overexpression in a relatively high percentage of patients, it seems that p53 mutation plays an important role in development of colorectal carcinoma. There was no significant association between p53 protein expression and some common clinicopathologic variables such as age, gender, site of tumor, pathologic type, and stage of the disease.

Abstract:

Background: Wilms' tumor is an emberyonal tumor arising from remnants of immature renal tissue. Her2/neu is an onco-protein which mediates cellular proliferation, differentiation and survival. Methods: In the current study, we analyzed Her2/neu expression in 40 Wilms' tumors. The clinico-demographic data of 40 patients with Wilms' tumor were retrieved. Immunohistochemical staining for HER2/neu was performed.Her2/neuimmunoreactivity was evaluated by Canadian Consensus 2007 scoring system. Results: Among the 38 specimens with epithelial component, 68.5% were positive for Her2/neu, whereas there was immunoreactivity in 37% of 38 blastemal, and 12% of 31 stromal components. The Her2/neu expression was significantly higher in early stages (81.5%) than in advanced stages (36.4%) in epithelial component, but not in other components. Conclusion: This study suggested that Her2/neuexpression is associated with epithelial cell differentiation accompanied by lower stages of tumor. No significant relationship was found between Her2/neu positivity and tumor size and patient's age and gender.

Abstract:

Background: The increase in high-risk behaviors (HRBs) is recognized as a serious threat to the health of societies. Self-expression is one of the social skills that can have a positive effect on controlling some risky behaviors. Objectives: This study aimed to investigate the correlation between self-expression and HRBs among newly arrived students at Isfahan University of Medical Sciences. Methods: In this cross-sectional study, 402 students (166 males and 236 females) were recruited from Isfahan University of Medical Sciences in 2020. They were selected using a multi-stage random sample and answered questions from the self-expression questionnaire and the high-risk behavior questionnaire. Various statistical indicators were utilized, such as mean, standard deviation (SD), t-test for comparison between the two gender groups, ANOVA for the mean difference between three age groups, and a Pearson correlation matrix to examine the correlations between all components of HRBs. Additionally, the Pearson test and regression analysis were employed. Results: A significant negative correlation was found between the self-expression score and HRBs (r = -0.56, P < 0.001). The mean score of HRBs was 15.33 ± 18.047. The mean score of self-expressions in male students was significantly lower than that of female students (74.51 ± 16.33 vs. 80.36 ± 14.30). The most common HRBs were related to smoking, hookah use (mean = 7.53 ± 8.75), and suicidal ideation (mean = 3.11 ± 3.93). Conclusions:It accentuates the significance of incorporating self-expression interventions in educational and health programs to foster healthier behaviors and promote mental health among this vulnerable population. Further research is warranted to explore additional factors influencing the link between self-expression and HRBs among university students.

Abstract:

Background: Gastric and colorectal adenocarcinoma are the second and the fifth most common cancers in Iran, respectively. Expression of cytokeratin 7 (CK7) is established in most malignancies including gastric and colorectal adenocarcinoma. Demonstration of Ck7 could be related to prognostic factors and help to the better management of the patients. The objective of our study was to evaluate the CK7 expression in gastric and colorectal adenocarcinoma and its correlation with other prognostic factors. Methods: This cross-sectional study was performed on 99 tissue blocks from patients with gastric or colorectal adenocarcinoma undergoing surgery. Tumor grade, tumor size, depth of invasion and metastasis to lymph nodes were determined. Then, the expression of CK7 was studied using immunohistochemistry staining. Results: Expression of CK7 was 50% and 33.8% in gastric and colorectal adenocarcinoma, respectively. There was not only a significant correlation between CK7 expression and tumor size (r=0.267, P=0.009) but also histologic grade (r=0.222, P=0.028).Conclusion: CK7 could be more expressed with the increase in tumor size and was associated with poorly differentiated gastric and colorectal adenocarcinoma. However, with these results gathered, it is highly recommended that further studies will be conducted to reveal the exact prognostic role of this factor.
